Beneficial effect of mercury-induced acute renal failure


Abstract
This study was undertaken to determine whether hydroxyl radicals are involved in mercury chloride-induced acute renal failure in rabbits. Rabbits received subcutaneously HgClz in a dose of 10 mg/kg and alterations in renal funaction were measured 24 hr after mercury injection. The hydroxyl radical scavenger dimethylthiourea (DMTU) was given intraperitoneally in a dose of 500 mg/kg just before HgClz injection. Administration of HgClz caused a significant increase in serum creatinine level, a reduction in glomerular filtration rate, and an increase in fraction excretion of Na and K, indicating induction of acute renal failure. These effects were prevented by DMTU pretreatment. DMTU attenuated HgClz-induced impairment in proximal tubular reabsorption of glucose and inorganic phosphate. Prevention of renal dysfunction by DMTU was not associated with alterations in renal blood flow. These results suggest that HgClz-induced acute renal failure is associated with generation of hydroxyl radicals in rabbits.
